    I have a couple of questions: what’s the Glucose concentration that you used for low glucose? Do you see time-dependent adaptations to low glucose? I mean, how long after putting your cells in low glucose did you see changes in gene expression? Are this maintained if you keep the cells in low glucose or are they only an acute response? Finally, have you look whether there is any difference in glucose dependency in cells grown in 3D compared to 2D?

  • Hi Elena, thanks for the attention.
    For low glucose we used a medium with 5,5 mM of glucose, this concentration is actually in the range of the normal glucose concentration presented in healthy individuals, representing about 100 mg/dL. Some articles call this medium a Normal glucose medium as well. In this initial experiment, we only cultivated the cells for 24 hours in different glucose conditions, looking to evaluate the acute effect of modifications of glucose in the parameters evaluated. We now are starting the 3D cultivation to the comparison, I hope to have new results pretty soon to share with you.
